    Clear 3D Cell Culture Models With Minimal Changes to Morphology and Without Compromising Sensitivity

    Despite the growing relevance of 3D cell culture as a research model, imaging techniques used to characterize these models are highly limited. Due to the thickness and opacity of 3D cellular structures, most current imaging technologies cannot penetrate to the center of the tissues, resulting in only the outer 2 to 3 layers of cells being detected. This causes the dark centers often seen in images of 3D cell culture models, which proves highly problematic for accurate analysis as these outer cells are most exposed to compounds, nutrients and oxygen and thus do not reflect the entire cell population.

    Corning now offers 3D Clear Tissue Clearing reagent that can be used in a tissue clearing technique, designed specifically to support imaging for 3D cell culture models and plate-based high throughput processing. When paired with fluorescent labeling (e.g., fluorescent protein, immunofluorescence, chemical dyes) and high content confocal microscopy, Corning 3D Clear reagent allows for complete 3D cell culture model characterization and more accurate drug screening.

    With L-glutamine Leibovitz’s L15 medium was originally developed without a sodium bicaronate buffer system for use in non-CO2 equilibrated environments.  It is buffered by phosphates and the amino acid L-arginine, along with L-histidine and L-cysteine.  The formation of acid metabolite byproducts is prevented by replacing glucose with galactose and sodium pyruvate.  Examples of usage include the propagation of HEP-2, monkey kidney cells and primary explants of embryonic and adult human tissue.  Leibovitz’s L-15 supports established cell lines as well as primary explants of embryonic and adult human tissue.

    Growth Promotion and Cytotoxicity
    Corning classical media products undergo a variety of tests to determine functionality and toxicity using growth-promotion analysis. Product performance is measured by harvest-to-plant ratios or fold increase according to predetermined specifications.

    Physiochemical Properties
    Liquid and powder classical media products are tested to determine pH and osmolality. Classical media powders are also tested for residual moisture. The pH and osmolality tests are performed according to specific procedures in which all equipment is calibrated using standards traceable to the National Institute of Standards and Technology.

    Biological Tests
    Quality release testing of Corning liquid classical media products assures low levels of endotoxin, mycoplasma, and an absence of contamination.

    Endotoxin
    Classical media products are tested for bacterial endotoxin using the Limulus Amoebocyte Lysate (LAL) chromogenic assay. For release, each lot of standard liquid classical media must have an endotoxin level of <0.25 EU/mL. Classical media powder products must have an endotoxin level of <0.25 EU/mL when reconstituted to formula weight with low endotoxin water.

    Sterility
    All Corning liquid classical media products are tested for sterility.

    Mycoplasma
    Liquid classical media products are tested for mycoplasma using a large-volume method.

    Heavy duty bottle which can be used for storage as well as mixing and sampling. Includes linerless, one-piece autoclavable polypropylene plug seal cap with drip free pouring rings. Optional red PBT caps and red ETFE pouring rings are available for dry heat sterilization (180°C). Glass bead indicates full capacity line on 100 mL through 2L size. Bottles have permanent white enamel graduations and marking spots. Color-coded polypropylene caps are available in a choice of orange, green, purple, red, and light gray.

    Caution: Bottles larger than 2L should NOT be used with bottle top filter units, or in other applications involving vacuum pressure, as breakage may occur. DO NOT have caps tightened immediately after autoclaving, as the vacuum resulting from cooling can cause breakage. A pouring ring is not included on 25 mL size. A vented cap is NOT recommended for use on the 5L and 10L 1395 series bottle.
